IL-2 IS stands for interleukin 2 “Improved Sequence”, also termed aldesleukin, and is a variant of IL-2 with a serine to cysteine substitution. Human IL-2 IS is a recombinant protein optimized for use in cell culture, differentiation studies, and functional assays, and possesses the same properties as naturally occurring IL-2.

Background information

IL-2, a potent lymphoid cell growth factor, is a typical four α-helix bundle cytokine. It is produced by activated T cells, especially the CD4+ T helper cell population. It plays an important role in both the activation and maintenance of immune responses and in lymphocyte development. IL-2 promotes proliferation and differentiation of T cells, NK cells and B cells and is involved in the elimination of self-reactive T cells. IL-2 signals through a receptor complex consisting of IL-2 receptor α-chain (CD25), β- chain, and common γ-chain. The latter two are also used for IL-15 signaling.

Quality description

Research-grade cytokines are suitable for a wide variety of cell culture applications. They are sterile-filtered prior to lyophilization. Generally, endotoxin levels are <0.1 ng/μg (<1 EU/μg), and purities are >95%. The biological activity is tested in appropriate bioassays.

Premium-grade cytokines offer the convenience of high and well-defined biological activities and allow exact unit dosing for demanding applications. The biological activity is determined after lyophilization and reconstitution, and normalized to WHO/NIBSC standards whenever available. In general, endotoxin levels are <0.01 ng/μg (<0.1 EU/μg), and purities are >97%. Lot-specific certificates of analysis are available on request (macstec@miltenyibiotec.de).

BACKGROUND
Interleukin 17 （also known as CTLA8）
is a T cellexpressed
pleiotropic cytokine that exhibits a high degree of homology to a protein encoded by the ORF13 gene of
herpesvirus Saimiri. cDNA clones encoding IL17
have been isolated from activated rat, mouse and human T cells. Human IL17
cDNA encodes a 155 amino acid （aa）
residue precursor protein with a 19 amino acid residue signal peptide that is cleaved to yield the 136 aa residue mature IL17
containing one potential Nlinked
glycosylation site. Both recombinant and natural IL17
have been shown to exist as disulfidelinked
homodimers. At the amino acid level, human IL17
shows 72% and
63% sequence identity with herpesvirus and rat IL17,
respectively. An IL17
specific mouse cell surface receptor （IL17
R） has been cloned. While the expression of
IL17
mRNA is restricted to activated T cells, the expression of mIL17
R mRNA has been detected in virtually all cells and tissues tested. IL17
exhibits multiple
biological activities on a variety of cells including the induction of IL6
and IL8
production in fibroblasts, the enhancement of surface expression of ICAM1
in
fibroblasts, activation of NFκB
and costimulation of T cell proliferation. The nonglycosylated,
E. coliexpressed
rhIL17
has been shown to be active on human as
well as mouse cell lines.

General Information
Interleukin-33 （IL-33） is a recently identified member of the IL-1 family of cytokines whose other members include IL-1-a/beta, IL-1Ra and IL-18. Its receptor has been shown to be ST2, an IL-1 receptor family member that also acts as a negative regulator of TLR-IL-1R signaling and IL-1R accessory protein （IL-1RAcP）. Receptor binding of IL-33 activates NF-kappa-B and MAP kinases and induces the expression of TH2-associated cytokines such as IL-4, IL-5 and IL-6. Prolonged IL-33 treatment of mice led to the development of eosinophilia, splenomegaly, and severe pathological changes in mucosal organs such as lungs, esophagus and small intestine. Recent experiments have shown that IL-33 can also co-localize with heterochromatin and possesses transcriptional repressor activities, indicating that IL-33 may function as both a proinflammatory cytokine and an intracellular nuclear factor with transcriptional regulatory properties.
