Background information
CD25, the low-affinity interleukin-2 receptor alpha chain (IL-2Rα), is expressed on activated T and B cells and monocytes. CD25 is also present on a subset of non-activated CD4+ T cells, which has regulatory function.
The CD25 MicroBeads II recognize epitope A of the CD25 molecule.
For control staining of cells separated with CD25 MicroBeads II, it is recommended to use MACS® CD25 Antibodies, which bind to epitope B of the CD25 antigen.
For isolation of CD25+ cell subsets, the CD25 MicroBeads II can be combined with MACS® Cell Isolation Kits or MultiSort Kits. The CD4+CD25+ Regulatory T Cell Isolation Kit is recommended for most efficient separation of CD4+CD25+ regulatory T cells.

CD138 MicroBeads were developed for the positive selection or depletion of plasma cells from PBMCs, bone marrow, leukapheresis harvests, and single-cell suspensions from lymphoid tissues.
0.2 µm filtered in phosphate-buffered solution, pH 7.2, containing no preservative. Endotoxin level is <0.1 EU/µg of the protein (<0.01 ng/µg of the protein) as determined by the LAL test.
Preparation:
The LEAF™ (Low Endotoxin, Azide-Free) antibody was purified by affinity chromatography.
The SK7 (Leu-4) monoclonal antibody specifically binds to the epsilon chain of the CD3 antigen/T-cell antigen receptor (TCR) complex. This complex is composed of at least six proteins that range in molecular weight from 20 to 30 kDa. The antigen recognized by CD3 antibodies is noncovalently associated with either α/β or γ/δ TCR (70 to 90 kDa). The CD3 antigen is present on 61% to 85% of normal peripheral blood lymphocytes 60% to 85% of thymocytes and on Purkinje cells in cerebellum. The soluble form of this antibody has a mitogenic effect on most peripheral blood T lymphocytes, provided appropriate functional monocytes are present.

PE-Cy™7 is a tandem fluorochrome that combines PE and a cyanine dye. PE-Cy7 conjugated reagents are as bright as PE conjugates. PE-Cy7 is particularly sensitive to photo-induced degradation, resulting in loss of fluorescence and changes in fluorescence spillover. Extreme caution must be taken to avoid light exposure and prolonged exposure to paraformaldehyde fixative. Fixed cells should be analyzed within 4 hours of fixation in paraformaldehyde or transferred to a paraformaldehyde-free buffer for overnight storage.
The RPA-T4 monoclonal antibody specifically binds to CD4, a 59 kDa single-chain transmembrane glycoprotein that is expressed on T-helper/inducer cell populations. CD4 is also expressed on thymocytes subsets and at lower levels on monocytes and macrophages. CD4 functions as a co-receptor in MHC class II-restricted antigen-induced T cell activation and as a receptor for human immunodeficiency viruses (HIV). This antibody binds to the D1 domain (CDR1 and CDR3 epitopes) of the CD4 antigen and reacts with approximay 80% of thymocytes and 45% of peripheral blood lymphocytes. RPA-T4 is capable of blocking HIV-1, gp120, and inhibits syncytium formation.
This antibody is routinely tested by flow cytometric analysis. Other applications were tested at BD Biosciences Pharmingen during antibody development or are reported in the literature.

R-phycoerythrin (PE) is an accessory photosynthetic pigment found in red algae. It exists in vitro as a 240-kDa protein with 23 phycoerythrobilin chromophores per molecule. This makes PE one of the brightest fluorochromes for flow cytometry applications, but its photobleaching properties make it unsuitable for fluorescence microscopy.

DESCRIPTION
The RPA-T8 monoclonal antibody specifically binds to CD8 alpha (CD8α) . CD8α is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily. CD8α is expressed by the majority of thymocytes, by subpopulations of αβ T cells and γδ T cells and by some NK cells. Cell surface CD8α is expressed either as a disulfide-linked homodimer (CD8αα) or as a heterodimer (CD8αβ) when disulfide-bonded to a CD8 beta chain (CD8β) . CD8-positive αβ T cells coexpress both CD8αα homodimers and CD8αβ heterodimers whereas some γδ T cells and NK cells express CD8αα homodimers. CD8 plays important roles in T cell activation and selection. The extracellular IgSF domain of CD8 α binds to a non-polymorphic determinant on HLA class I molecules (α3 domain) and enables CD8 to function as a co-receptor with MHC class I-restricted TCR during T cell recognition of antigen. The cytoplasmic domain of CD8α associates with Lck, a Src family protein tyrosine kinase that is involved in intracellular signaling. The RPA-T8 and HIT8a monoclonal antibodies are not cross-blocking. This clone has been reported to react with a subset of peripheral blood lymphocytes, but not monocytes nor granuloyctes, of baboon and both rhesus and cynomolgus macaque monkey. In general, a higher frequency of CD8+ and CD4+CD8+ lymphocytes are observed in non-human primates compared to normal human donors.

FITC, fluorescein isothiocyanate, is a fluorochrome with a molecular weight of 389 Da. FITC is sensitive to pH changes and photobleaching. Due to nearly identical excitation and emission properties but different spillover characteristics, FITC and Alexa Fluor® 488 cannot be used simultaneously. FITC is relatively dim and should be reserved for highly expressed markers whenever possible.

DESCRIPTION
The 3G8 monoclonal antibody specifically binds to the 50-65 kDa transmembrane form of the IgG Fc Receptor (FcγRIII), a human NK cell-associated antigen. CD16 is expressed on NK cells as well as macrophages and granulocytes. Reports indicate that CD16 plays a role in signal transduction and NK cell activation. The 3G8 antibody blocks the binding of soluble immune complexes to granulocytes. The 3G8 antibody is reported (Vossebeld et al., 1997) to increase intracellular calcium levels in human neutrophils by interacting with bothFcγRIIa and FcγRIIIb molecules. This antibody has also been reported to induce homotypic neutrophil aggregation.

Peripheral T-cell lymphoma. The majority of the neoplastic cells show a strong distinct membranous staining reaction.
Clinical application for IR649/IS649. For identification of thymocytes, T-helper cells and classification of related tumors such as peripheral T-cell lymphoma (unspecified), anaplastic large cell lymphoma and mycosis fungoides.
CD8 is a 68 kDa transmembrane glycoprotein expressed as a heterodimer by a majority of thymocytes, and by class I major histocompatibility complex restricted, mature, suppressor/cytotoxic T cells.
The antibody is a useful aid for classification of cytotoxic/suppressor T-cell lymphomas.
CD33 antigen is reported to appear on myelomonocytic precursor cells after CD34 antigen expression. It then continues to be expressed on both the myeloid and monocyte lineages, although it is reported to be absent on granulocytes. It has been reported that expression of CD33 is restricted to monocytes, premyelocytes, myeloid blasts, some acute undifferentiated leukemias and acute lymphoblastic leukemias.
