DTT is synthetic. A method of preparation has been described in J. Chem. Soc. 253 (1949)
DTT is an agent for reducing -S-S- to -SH. Redox potential: -0.33 volts at pH 7. DTT acts as a protective agent for -SH groups of other species in solution. In this reaction the DTT is oxidized to the cyclic disulfide, and thereby ensures the reduction of other disulfides in solution. The disulfide reduction is complete in minutes at pH 8. A specific and sensitive assay for disulfide groups is based on determination of resulting monothiols with DTNB (Fluka 43760).2
Applications:
It is used in buffers for isolation of RNA5 or buffers used for RT-PCR starting with mRNA6,7.

Preparation Instructions:
The solubility of DTT has been tested by Fluka at 50 mg/ml in water, resulting in a clear colorless solution. It has been described as freely soluble in water, ethanol, acetone, ethylate, chloroform and ether.
Solutions oxidize relatively slowly in air.3 It is recommended to make solutions fresh daily. Recorded half-life (hours) of DTT solutions at various pH and temperatures are as follows:4

Principle

Effectene Transfection Reagent is an innovative non-liposomal lipid formulation that is used in conjunction with a special DNA-condensing enhancer and optimized buffer to achieve high transfection efficiencies. The enhancer first condenses the DNA molecules and Effectene Reagent subsequently coats them with cationic lipids (see figure “Model of Effectene Principle”), providing a particularly efficient way of transferring DNA into eukaryotic cells. Effectene Reagent offers significant advantages over many liposome reagents and other transfection methods (1, 2).

Procedure

The Effectene procedure has two steps. DNA is first mixed with Enhancer and a buffer that provides optimal salt conditions for efficient DNA condensation. This step requires just 2–5 minutes. Effectene Reagent is then added and the mixture is incubated for 5–10 minutes to allow Effectene–DNA complexes to form. The complexes are mixed with growth medium (which can contain serum and antibiotics), and added directly to the cells. The cells are then incubated until harvested and analyzed for gene expression (see flowchart “Effectene Transfection Reagent”).

Applications

Effectene Reagent is suitable for transient and stable transfection of a broad range of cell types (see figures “High Transfection Efficiencies Using Effectene Reagent” and “Transfection of Oligonucleotides Using Effectene Reagent”). Because transfection can be performed in the presence of serum and requires low amounts of DNA (see figure “Serum and DNA Quantity vs. Transfection Efficiency”), cytotoxicity is minimal. This makes Effectene Reagent particularly effective for primary cells (see figure “Effectene Reagent with Primary Cells”). High-throughput transfection

The application of recombinant DNA technology to fields such as drug discovery and development has led to an increased need for high-throughput transfection. Transfection using Effectene Reagent requires low amounts of DNA and minimal handling, and in addition, removal of transfection complexes is not required, making this reagent highly suitable for high-throughput screening. Effectene Reagent provides outstanding transfection efficiencies, excellent reproducibility, and minimal cytotoxicity in high-throughput transfection, and is available in bulk quantities.

  注意事项:
 
  1、解冻血清时,请按照所建议的逐步解冻法(-20℃至4℃至室温),若血清解冻时改变的温度太大(如-20℃至37℃),实验显示非常容易产生沉淀物。
 
  2、解冻血清时,请随时将之摇晃均匀,使温度及成分均一,减少沉淀的发生.
 
  3、请勿将血清置于37℃太久。若在37℃放置太久,血清会变得混浊,同时血清中许多较不稳定的成分也会因此受到损害,而影响血清的质量。
 
  4、血清的热灭活非常容易造成沉淀物的增多,若非必要,可以无须做此步骤。
 
  5、若必须做血清的热灭活,请遵守56℃,30分钟的原则,并且随时摇 晃均匀。温度过高,时间过久或摇晃不均匀,都会造成沉淀物的增多。

Specificity Ubi-1 was initially screened on ELISA of the immunogen, and subsequently tested on sections of Alzheimer brain. It was one of several
clones which stained neurofibrillary tangles in frozen sectioned material strongly and specifically. Subsequent studies indicated that Ubi-
1 is relatively insensitive to formalin fixation and so can be used on mildly fixed histological sections of human brain for studies of
Alzheimer's disease. Ubi-1 also recognizes other ubiquitinated inclusion bodies such as the Lewy bodies of Parkinson's disease and
the Pick bodies in Pick's disease. This antibody recognizes polyubiquitin chains much more strongly than monoubiquitinated molecules
or free ubiquitin. Specifically recognizes ubiquitinated cytoplasmic inclusion bodies.

Target
Relevance Ubiquitin is a highly conserved protein of about 8.5 kDa molecular weight, which has an ATP dependent role in the targeting of proteins
for proteolytic degradation. To perform this function, the protein to be degraded is first covalently attached to the C terminus of ubiquitin,
and the ubiquitinated complex is then recognized by a complex of degradative enzymes. Interestingly, ubiquitin also becomes covalently
bonded to many types of pathological inclusions, which appear to be resistant to normal degradation. Therefore, ubiquitin antibodies
are very useful for studies of these inclusions. For example, the neurofibrillary tangles and paired helical filaments diagnostic of
Alzheimer's disease, Lewy bodies seen in Parkinson's disease, and Pick bodies found in Pick's disease are all heavily ubiquitinated
and can be readily visualized with ubiquitin antibodies.

5. Allow ImmEdge™ Pen reagent to dry before immersing slides in aqueous solutions.
