SPiDER-βGal
利用活细胞分析lacZ报告基因的表达
SPiDER-βGal
SPiDER-βGal是检测具有细胞膜透过性及细胞内滞留性β-galactosidase的新型试剂。SPiDER-βGal具有较高的细胞膜透过性,因此可使用活细胞分析分析lacZ报告基因的表达。色素保留在细胞内,不会向外流出。因而,可进行单细胞层面的高灵敏度成像。
◆原理
SPiDER-βGal与β-galactosidase发生反应,生成名为quinone methide(醌甲基化物)的中间体,与附近的蛋白质中的亲核基团(SH基等)结合,形成稳定的共价键,从而具备荧光性质。
反应后的色素被固定在细胞内的蛋白质,滞留在胞内,可在单个细胞层面检测β-galactosidase表达细胞。
◆特点
● 可使用活细胞或组织进行单个细胞层面的表达细胞荧光成像,亦可使用X-gal等固定细胞
● 为探测物不向外流出,使用比以往更高灵敏度的β-galactosidase的荧光探针进行检测

◆利用SPiDER-βGal检测β-galactosidase
SPiDER-βgal可对固定细胞及活细胞进行染色,即使染色后进行固定化,也不会对色图产生影响。
◆固定化对染色的影响
细胞固定化会使GFP的荧光强度下降。但是,使用SPiDER-βgal进行染色,即使是固定细胞也具有较强的荧光性。

果蝇组织的Live成像
用10 μmol/l SPiDER-βGal 和16 μmol/l Hoechst 33342对未固定的果蝇组织染色20-30分钟,共聚焦显微镜观察。

利用SPiDER-βGal在细胞内的高保留性,完全不会渗出到邻近细胞;只对β-galactosidase(β-半乳糖苷酶)表达细胞进行单细胞层面染色。

◆原理
KRN7000 是人和小鼠自然杀伤T(NKT)细胞的配体,在各种小鼠体内实验模型中,比如皮下移植模型、肝脏或者肺的转移模型等,KRN7000可刺激产生干扰素 (IFN),起着抗肿瘤活性的作用。在肝脏转移模型中,使用KRN7000治疗后,能够抑制肿瘤生长,延长小鼠寿命。据报道,在患有传染病,自身免疫性疾病和移植物抗宿主病等小鼠身上使用KRN7000后,会产生免疫效果。
自然杀伤T细胞(NKT细胞)被激活后会分泌大量的细胞因子,并能分化成具有细胞毒作用的效应细胞。临床研究发现NKT细胞参与部分疾病的调节,尤其是参与了自身免疫病,抗肿瘤和器官移植的抗排斥反应的调节。特别是NKT细胞在一些自身免疫性疾病如硬皮病,I型糖尿病及肝炎等具有免疫保护作用。
目前已知对NKT细胞的刺激效应最佳的物质是脑苷脂-α-半乳糖酰基鞘胺醇(α-Galactosyl cermaide,α-GalCer,KRN7000),这类物质来源于海绵动物。1993年由日本Kyowa Hakko Kirin公司首次合成并命名。尽管KRN7000对NKT细胞的天然调节作用不明,但它作为一种NKT细胞强有力的活化剂,在体内体外实验中非常有用,NKT细胞通过这种机制活化后,随后会激活NK细胞。

体内注射:KRN7000可溶于以下溶液:5.6% 蔗糖,0.75% L-组氨酸,0.5% 吐温-20。混合后,加热至80℃,直至KRN7000完全溶解。将溶液冻干后,冻干粉易溶于水。
体外实验:KRN7000可溶于DMSO,终浓度为1mg/mL,加热至80℃得到澄清溶液。
案例1
使用KRN7000用于鉴定小鼠组织或者人血液样品中iNKT细胞(CD1d限制性恒定NKT细胞)。iNKT细胞受体由小鼠中恒定的Va14Ja18/Vb8.2和人Va24Ja18/Vb11组成。这两种物种的iNKT细胞抗原受体都识别MHC I样CD1d递呈的a-galactosylceramide (a-GalCer,KRN7000)。使用流式检测a-GalCer装置的CD1d二聚体即可进行分析。从5~6只小鼠胸腺,脾脏或者肝脏可以纯化1×106 iNKT细胞,大概需要1~2小时从小鼠组织制备单核细胞,用磁珠大概1.5小时富集细胞,4小时用流式检测和纯化iNKT细胞。如果样品是人的PBMCs(外周血单核细胞),大概需要2小时,需要5mL血液获得5×106PBMCs,含有500~25,000 iNKT细胞。

案例2
肉状瘤病是系统失调疾病,可能与Th1应激有关。在患有non-remitting肉状瘤病病人外周血中Va24 NKT细胞数目减少,但是在肉芽肿病灶部位这些细胞有积累。用KRN7000刺激来自上述病人外周血的Va24 NKT细胞,IFN-gamma的分泌量明显比正常人少,但是IL-4的水平差不多。只有在Va24 NKT中观察到IFN-gamma分泌减少,在常规的CN4 T细胞中没有变化。表明non-remitting 肉状瘤病与Va24 NKT 不能分泌足够IFN-gamma有关,同时也说明Va24 NKT细胞在肉状瘤病的病理有关。
